I know it had some spirits added to Smash back when it debuted but given how immensely popular the game has been, the longer (pandemic-induced) development cycle for Season 2 that could potentially result in plans changing, and the upcoming anniversary, I would be surprised if Animal Crossing doesn't get a New Horizons adjacent rep in Ultimate at least before the season's out.

Isabelle was announced alongside the new Animal Crossing, yes. But given how huge New Horizons has been after its release and the longer development cycle for Season 2 due to you-know-what I would not be surprised if Nintendo demanded requested that Sakurai add another Animal Crossing fighter.
I mean, Smash Bros. is all about making Nintendo money, and I could see them wanting another new Animal Crossing character in Smash now to cash in on New Horizons' popularity.

A number of characters already in Smash didn't exactly scream "fighting game character" before they got in, either. And "There's already been a new one" does not mean there can't be another new one.
Maybe it'll just be some Mii Fighter skins. Maybe Isabelle and Villager will get a new pack of alt skins (not that anybody has yet). But I'm still not about to write off the possibility of Nintendo's biggest game of 2020 getting some additional representation in Smash.

Most of those non fighting game characters had something resembling an attack in their game though. I'd class 11 out of the 78 as characters who don't have any game based attacks, due to being cut scene based (Daisy and Sheik) or not being in an action game at all (Dr Mario, Wii Fit Trainer or Captain Falcon), so it's not a unique thing to Animal Crossing, and if they can come up with a way to add someone with a move set that isn't basically just Villager again I'll be fine with it. But with Villager and Isabelle covered, who would you pick? Nook is the only obvious choice, they can't pick a villager you have a minuscule chance of having. What on earth would his moves be? Final Smash: The Crippling Mortgage.
I think there are series which have interesting characters which would be more worthy of representation and could give new mechanics. Mike Jones for one. The last real new Zelda character was in Melee, so that's 20 years ago. Ryu Hyabusa would be cool. That's without going outside of NES era games.
Edit: or Radd Spencer. A Smash Bros character who couldn't jump could be fascinating

I could definitely see Tom Nook definitely getting a spot, but I disagree with this statement. It's not about villagers people have, it's about villagers people want. If Raymond or Audie got in Smash and then brought the other cats or wolves (both very popular villager species) along with them as alternate skins the fans would frickin' cheer.

Build furniture to chuck at the opponent. Have it be kinda like a charge attack; the longer the build-up the deadlier the projectile. Implement the vaulting pole and extendable ladder into the standard moveset. Have the Final Smash be building the Robot Hero to smite your enemies with.

The next Smash fighter is... all 391 Animal Crossing villagers. That's right, EVERYONE is here! For the most part they share a single moveset, with small variations (a la Echo Fighters) for each of the eight personality types. Each player who downloads the new fighter will be assigned a villager at random. Don't like who you got? Lose enough matches with them to lower your friendship level, and there's a chance they'll move out of your roster and be replaced with someone else; you must defeat the new arrival to convince them to move in. Got Raymond? Congratulations, your copy of Smash is now worth five hundred dollars.
What if they remove someone from Smash instead? Change the meta or something?

I want to be playing No More Heroes 3 tonight, Nintendo.
Actual for real for real I bet we get a Mother collection for Switch, including an English translation of Mother 3. Earthbound is such a massive hole in the Switch Online SNES collection they have to be holding it back for some reason. It seems like such an easy home run at a time when larger scale game development seems to be suffering because of the pandemic.

For my realistic, non-Smash predictions, they been promoting Genealogy of a Holy War characters a lot in FE Heroes lately, so I wonder if another FE Echoes games is in the works. Also, something for the Zelda anniversary, whatever it ends up being, seems likely. I’m not getting my hopes up for anything Metroid anniversary related.

Something Zelda is certain to show. Be it a collection of games, DLC for Age of Calamity or BotW 2. Metroid may well show in some form, Prime 4 was restarted 2 years ago and it's the anniversary. Maybe Bayonetta 3?

Best case scenario: More new (and/or old) villagers joining the roster, more new swag and returning major NPCs, Amiibo for the animal friends who don't have any yet. Really any of these.
Neutral case scenario: Just a showcase of the Mario swag we already know is coming in March.
Worst case scenario: Not a bloody thing featured in the Direct. Or some form of microtransactions.
